چکیده

An Escherichia coli gene which is able to complement "ammonia-dependent growth" (adgA) mutations in Rhodobacter capsulatus was previously isolated from an R-prime plasmid and was shown to be located between 34 and 39 min on the E. coli chromosome (3). In the present study, a 10.9-kb HindIII-PstI fragment of genomic DNA was cloned from E. coli HB101 and shown to complement adgA mutations in R. capsulatus. Restriction mapping of this fragment and comparison with the restriction map of the E. coli chromosome (5) (Fig. 1) showed that the complementing gene is located at 38.1 min (1840 kbp) on the E. coli genetic map. The restriction map of this fragment was identical to that of Kohara et al. (5), except for the presence of an additional EcoRV site. The chromosomal location of the gene is identical to that proposed by Kim and Wulff (4) on the basis of the partial sequence of an 8.0-kb DNA fragment. Restriction fragments containing the Kmr gene from pUC4-KIXX (Pharmacia-LKB) were inserted, in both orien-

Recycling of the posttermination complexes of Mycobacterium smegmatis and Escherichia coli ribosomes using heterologous factors.

In eubacteria, ribosome recycling factor (RRF) and elongation factor G (EFG) function together to dissociate posttermination ribosomal complexes. Earlier studies, using heterologous factors from Mycobacterium tuberculosis in Escherichia coli revealed that specific interactions between RRF and EFG are crucial for their function in ribosome recycling. Here, we used translation factors from E. col...

Study of Mutations in the DNA gyrase gyrA Gene of Escherichia coli

Quinolones are a large and widely consumed class of synthetic drugs. Expanded-spectrum quinolones, like ciprofloxacin are highly effective against Gram-negative bacteria, especially Escherichia coli. In E. coli the major target for quinolones is DNA gyrase. This enzyme is composed of two subunits, GyrA and GyrB encoding by gyrA and gyrB, respectively. Mutations in either of these genes cause qu...

Identification of Biofilm Encoding Genes (agg) in the Escherichia coli Isolates by Multiplex-PCR Method

Introduction: Biofilm is a collection of microbial cells that are irreversibly suppressed and mildly washed away. The aggR gene is located on the main plasmid (pAA), which codes for many actuator factors. The pathogen strains of EAEC have an aggR gene, the gene has been identified in six classes of pathogenic E. coli.   Materials & Methods: In the present study, 60 stool samples from children ...

Presence of multi-drug resistant pathogenic Escherichia coli in Zarjob River located in The State of Guilan, Iran

In modern medical processes, new antimicrobial drugs are being widely used in order to rapid and essential eradication of resistant bacterial strains. Broad-spectrum beta-lactamases are enzymes originated from mutations in the common plasmid genes mediators for beta-lactamases such as TEM-1, TEM-2, and SHV-1 are transferred among bacterial species. The genes coding the production of ESBL are of...
